As this is a lab-based role, they will be responsible for developing and implementing a robust in-house mammalian expression platform. They will be accountable for consolidating and the running of the mammalian cell culture lab or related activities. In addition, they will also be expected to contribute to continuous improvement projects or other core activities beyond mammalian expression technologies within Constructive Bio. Responsibilities include: Establishing an optimised, robust and high yielding transient expression platform for recombinant protein production in HEK293 and CHO cell lines. Vector engineering and optimisation to enhance protein expression. Continuous review of processes, troubleshooting and implementation of optimised processes. Evaluating or developing new technologies and investigating possible implementation of new protein expression strategies where necessary. Running and management of all cell culture activities including continuous process improvement. Designing and conducting experiments according to project requirements. Supporting and managing the general day-to-day laboratory activities within team. Drafting of appropriate reports as required. Implement and drive Constructive Bio’s health and safety practices including the regulations governing COSHH, biological agents and genetically modified organisms as detailed in relevant Constructive Bio risk assessments. Developing and running analytical assays such as protein quantitation, SDS-PAGE, CGE, Western blotting, cell-based assays according to project requirements. Additional Responsibilities: Contributing to other core activities and relevant projects within Constructive Bio including presentation at appropriate internal meetings Liaising with other groups within Constructive Bio where required to apply new developments to future projects. Maintaining a detailed and up to date documentation of experiments in a timely manner according to Constructive Bio’s lab book requirements. The requirements for this position are: BSc/MSc/PhD in Life Sciences plus extensive experience in cell culture and in establishing mammalian protein expression systems (especially HEK293 and CHO cell lines) preferably gained in an industrial environment. In depth experience of optimising and implementing transient protein expression in mammalian systems. Proven expertise in molecular biology and cell biology techniques. Experience in analytical techniques such as protein quantitation, SDS-PAGE, CGE, Western blotting. Ideally you will also demonstrate: Experience in cell-based assay development Familiarity or experience with alternative expression systems including microbial expression systems Experience in stable expression systems Behavioural competencies Independent, Initiative, proactive, good communicator, good team player, excellent scientific methodology.
