Location of role Oxford, UK Department Proteomics Key Responsibilities We have an opportunity in our Research Group for a 12-month placement (Year in industry). We are looking for enthusiastic and highly motivated individuals who would relish the chance to learn from Immunocore’s cutting edge science and corporate growth. A key role of the Proteomics department is to identify and validate peptides presented by HLA complexes (pHLA) to the immune system, i.e. the immunopeptidome. Quantification of immunopeptides is paramount to ensure the selection of the best targets for our ImmTAX immunotherapy. The multistep nature of the immunopeptidomics workflow can lead to peptide losses that are not accounted for using standard mass spectrometry approaches for absolute quantification. We aim to improve current methodologies for immunopeptide quantification and to assess losses at different stages of the immunopeptidomics workflow. We believe that the new workflow will help us better characterise and understand target expression and will have a positive impact on selection of future targets for immunotherapy. Key Responsibilities This project aims to assess and improve key steps in our immunopeptidomics workflow by: establishing methods for generating stable isotope labelled pHLA complexes (SILpHLA); using SILpHLA reagents to assess immunopeptide losses during immunopurification; perform absolute quantification of immunopeptides in cell lines; compare absolute quantification using SILpHLA reagents with our established absolute quantification workflow. The successful candidate will gain experience in: Generating novel reagents for peptide quantification Targeted proteomics/immunopeptidomics using state-of-the-art mass spectrometry Cell culture and aseptic technique We envision that this work will lead to improved protocols for absolute quantification of immunopeptides, helping us to better understand the expression of targets in cancer cell lines and tissues. Education & Qualifications Undertaking (currently in second year and looking for a year in industry) BSc. in Biological or Biomedical Sciences with an immunology component. What will you receive?
